When we cast a 10% SDS PAGE gel, the protein resolution is not sufficient and is similar to an 8% gel. We tried using a freshly prepared acrylamide from a different box, but the problem still persists. What can be done to troubleshoot this problem? Which other reagents can cause this?

Try going for 12% gel. Try freshly prepared APS along with acrylamide. What about the pH of the .5M and 1.5M tris? Check that as well. Most importantly prepare the fresh Running buffer and then see the results. Hope it helps

Thanks Vikrant. We tried using APS from other lab today and it worked fine. Turns out APS is the problem.
