what is the best way to lysis yeast cells such as pichia pastoris or saccharomyces? i used bead beater and french press and found that they both cause heat to denature the protein.
any mechanical lysis tends to heat up the sample so you have to adapt the condition to avoid it. To improve the yield of lysis you could prepare spheroplasts in the first move.
You could try a freeze thaw cycle (or multiple cycles). I use this for rupturing cells for DNA extraction. Suspend cells in a buffer and freeze in liquid N or -80C for an hour then thaw at room temperature
