sex-determining region of the human Y chromosome (SRY), can be used to differentiate male from female DNA. Deletions/mutations may cause sex reversal in humans.

SRY Forward 5Õ-GAATATTCCCGCTCTCCGGAG-3Õ,

SRY Reverse 5Õ-ACCTGTTGTCCAGTTGCACT-3Õ

the above primers can be used to amplify a 418 bp fragment.

I also have some recent experience using a pair of SRY gene primers for genetic sex determination in our patients with indeterminate sex. I used:

SRY forward 5'-tacaggccatgcacagagag-3', and

SRY reverse 5'-tcttgagtgtgtggctttcg-3'.

It results in 179 bp amplicon, which separates quite well in 2% agarose.

How did you get the DNA from the fetus for this analysis and what is the backround of your study?

Hi Sven....we use chorionic villi or amniotic fluid sample of the fetus to get DNA and amplify the required gene

If you are looking for a good set of primers that is specific for BOTH X and Y yielding PCR products of different sizes as a standard I use ZFY/ZFX or AMELGY/AMELGX.Both serve the purpose well and are quite sensitive.  If you need the sequences I would be happy to send them.
