I read an article where the Aluminum Chloride Method is used for the determination of total flavonoid content after the reaction between the sample and AlCl3 took place they add potassium acetate. I want to know why is necessary to add the acetate in this reaction and which role does it has?
I am sure of the first part of my answer but I am "guessing" about the second part.
The reason for using aluminium chloride in the colorimetric method is that AlCl3 (being weakly acidic) forms stable complexes with the C-4 keto groups and either the C-3 or C-5 hydroxyl group of flavones and flavonols. In addition, it also forms acid labile complexes with the orthodihydroxyl groups in the A- or B-ring of flavonoids.
Potassium acetate is a basic salt "it comes from the weak acetic acid & the strong KOH" so it is added to utilize its weak basicity to neutralize the weak acidity in the system.
