It would be helpful if you could provide some detail as to what you are trying to analyse and how you are extracting and purifying your polysacchaide(s). Nevertheless, I can say immediately that you will not be able to directly analyse any polysaccharides by GC. They are too large and polar and thus non-volatile. You can, however, analyse their constituent monosaccharides (eg hexoses and pentoses) by GC after appropriate derivatisation.

Your polysaccharides will need to be hydrolysed and the released monosaccharides derivatised. The methods to be used will depend on the polysaccharide(s).

It is very difficult to analyse polysaccharides or any carbohydrates by using GLC.

If you have HPLC system then go for it for analysis.

Otherwise in GLC analysis, first you hydrolyse your sample by using acid and convert your polysaccharides in monosaccharides and analyse on GLC after appropriate derivatisation.
