I have digested the polysaccharides with HCl. Can I inject digested content (with acid) into the LCMS column for detection of individual mono or disaccharides ?
Any suggestion or advice, those who are working on carbohydrates
I have worked with LCMS only a few times, but as I know it is not a good idea to inject corrosive acids (or salt solutions after neutralization) into this instrument, unless you can afford to buy a new one.
The question is whether LCMS is really necessary to answer your research question. Maybe HPLC is sufficient, too.
Alternatively, you can try to use a less corrosive and MS compliant acid like formic acid for the digestion.
I think, there is a person in charge for this instrument at your institution. Maybe s/he has got more ideas.
I don't know your exact research question, but in general, HPLC with RID is a good choice for the analysis of mono-, di-, trisaccharides. For this system, there are several columns available that separate sugars. Some of them use diluted H2SO4 as eluent. Then, it's not problem to inject acidic samples directly. Good luck.
