I am trying to extract exopolysaccharide from fungal broth. Before doing ethanol precipitation step, i want to reduce the volume of 3L fungal broth to 300 mL. Is there any method to achieve this other than boiling?
Dear Deepthi,
you can try evaporation under preesure using a rotatry evaporater. Please, adjust the device parameters at 90-100 mbar and 45-55 C for pressure and temperature, respectively. At these conditions, you can concentrate your broth withouth going at higher boiling temperatures.

Will 45-55 C can cause polysaccharide degradation? Is lyophilization a good method?
I think till 80 C, there is no problem.
Lyophilization of such large volumes will not remove all solvents and will damage your device.
