we use Nile blue, and for this there is a need of a microscope at immersion, with fluorescence filters (exc. 515 nm/ emis. 575 nm). We also use a laser scanner, spotting drops of bacteria on slides, for comparative quantification of intensities.
Normal light microscope will be enough to observe PHB granule stained with sudan black. Observe in oil immersion lense. Add a drop or 2 or glycerol to the growth medium(slant) of bacterium to enhance production of PHB. Stain when the culture age is 18-24 h for better results.
what I meant is to compare the production among different strains or the same strain at diferent timeing, and check the peak of production. Ralstonia express a depolymerase so all the PHB produced is bound to be degraded.
