Actually, I am doing embryo rescue experiment on wild melon fruit. I would like to ask about how to isolate the embryo from seed. I also practiced by myself but it took so much time. To finish ten seeds I need 1 hour and it is very easy to make injured the embryo. I did in in the cleaning bench with the microscope so that it is also difficult to do. Do you have any suggestions for me? Thank you so much for spending time on my question?
Interspecific hybridization via embryorescue, in vitro pollination, somatic hybridization and cytogenetics of protoplasts. Embryo-rescue and in vitro pollination are suitable in vitro techniques for production of hybrid embryos. These methods were tested and optimized for cucurbits. Protoplast culture is another valuable tool for biotechnological applications, such as somatic hybridization and genetic transformation. For details see following publication:
