Hello, what do you think of extraction of 1kg crude plant material directly into acetone? I wish to get out an alkaloid with a fatty tail, I wanted to use dichloromethane but we dont have this solvent and it is too toxic in large quantities so I want to choose an extraction solvent with a lower polarity than methanol, would acetone be good?

Acetone is also polar solvent ,extraction with it will result in complex mixture (polar and non polar components) which is very difficult to isolate,therefore start with non polar solvent like Hexane or cyclohexane or ether then increase its polarity by adding 1% ,3%, 5%,10%.... acetone or methanol. Also, ethanol may work well too. Since you have a kg of material, try some small scale experiments to see what works best for you.
To extract alkaloids use chloroform by long time shaking maceration. After that you have to prepare an acid-base-extraction to isolate most of alkaloids compounds.
Yes, acetone herbal some extracts are in therapeutic practices, it is more close to body physiology.
Thank you for the answers and especially column recommendations, yes I also thought chloroform should be close in properties to dichlromethane, but chloroform would perhaps not be safe in such large amounts
Acetone extract may have polar and non-polar compounds and it will give effective result in the Soxhlet extraction. After extraction, you can use column with non polar solvent followed by polar solvent
I would suggest a water immessble solvent such as ethyl acetate or methyl t-butyl ether these solvents r not as polar as acetone you will get a cleaner extract. Also you can wash the extract with 5 percent sodium bicarbonate to remove acetic component then with water and dry the extract with anhydrous magnesium sulfate for several hours and then filter it. You will have a much cleaner extract. I second the suggestion that you try it first on a small scale to see how it works. You can use the suggested columns on the residue after expelling the solvent.
Ethyl acetate is less polar than acetone and it is not miscible with water.will give cleaner extract and you can wash the extrat with 5% sodium bicarbonate to get red of acidic compound as I indicate above. Good luck
I've found information that the calculated log P value for most of these alkaloids that I'm trying to extract is Log P~5, so now I wonder should hexane be an even better solvent?
Hexane can evaporate easily, so that is not compatible to extraction.
I have a suggestion. Use methanol for your extraction. After evaporation of methanol, you should use acidic solution such as TFA for extraction of alkaloids from methanolic extraction plates. Again evaporate acidic solution and use chloroform for the extraction of acidic plate’s extraction.

we have crude extract of grape seeds, extracted in aceton:water:acetic acid(90:9.5:0.5). Now my question is which solvent will be apt to be use for making different dilutions from above crude extract, which will be subsequently used in antioxidant assays.
