My question is about the function of Histidine in the medium. I know there is a mutation in our pichia cell line and we need to transform our plasmid into the host cells and make it able to produce Histidine and grow in Histidine deficient medium. The thing i want to know is why cell line without plasmid can grow in YPD but not in BMGY ?? Is not because of the glucose in YPD medium? does it play a role like carbon source?
