Percoll Gradient for C. novyi Spore Purification?

Hi,

I am doing Percoll Gradient for C.novyi Spore purification, and my results are not consistent.

I do 70%/55% as my gradient.

I put about 3 mL of 70% Percoll, and above it, I layer about 6 mL of 55% Percoll suspension of C. novyi Spore.

I centrifuge it at 3500 rpm for 15 minutes.

The pellet size is inconsistent every time, and the amount of pellet after Percoll gradient centrifugation is very low compared to just centrifugation of C. Novyi spore from its culture media.

Does anyone have input on C.novyi Spore purification?
