I have coated my SeNPs with PEG and paclitaxel drug. It was characterized using UV, to check for coating by absorbance peaks.I absorbed a shift in the peak. Is there any possible explanation to this?
The SeNPs powder was dissolved in PEG solution in 1:1 ratio and concentration maintained was 1mg/ml. The drug was added to the SeNPs-PEG solution and UV readings were taken.

In fact PEG is used to make selenium oxide nanoparticles. You need to have some sort of a coat either in the form of dots or a thin layer on these particles before you load a drug like paclitaxel. The oxide formation is just not enough to load the drug as it has a comparatively weak electronegativity.
1. Get an absorption spectrum of the plain SeNP, without drug, just to make sure what peaks are present.
2. You can also vary the amount of paclitaxel and see if the peak height changes accordingly; this would be hint that the peak you see is really due to the drug.
3. Don't know about nanoparticles, but Se is highly toxic by itself. Please keep this in mind.
1.If you perform UV do the studies individually (pure paclitaxel, SeNps with out drug and in SeNPs loaded with drug).
2. Or you can better opt for some other specific characterization of your coating process of SeNps.
