Hello,

I am generating droplets with microfluidics devices using soft lithography technique; the microchannel is 5 um width and 5 um depth.

However, I found a problem when I try to test with a needle and DI water.

The inlet and outlet ports were punched correctly.

When I observe in a microscope, the flow was stuck at the contact position between PDMS and glass slide. The ports occurs a backflow and the devices don't have any movement in the microchannel. (I handle with about 20 samples)

My laboratory is typical; it means no clean room.

In my opinion, the design of mold is good, but I am worried about the depth of it.

Any recommendations on how I can figure out my problem or tips, tricks or related article?
