Amyotrophic lateral sclerosis (ALS) affects both upper and lower motor neurons. TDP-43 pathology is observed in the majority of patients with familial or sporadic ALS (95%). The soma of both upper and lower motor neurons in the brain and spinal cord, respectively, are surrounded by the cerebrospinal fluid.

However, the levels of pathological TDP-43 proteins measured by ELISA are reported to be much higher in the plasma than those in the CSF.
