I am trying to isolate specific components from the E. coli biofilm extracellular matrix (which contains cellulose). I do not want to lyse cells, only break up the biofilm matrix.

Our lab has cellulase (powder) from Aspergillus niger with a concentration of 1.24U/mg. I'm struggling to find a concentration recommendation and the best solution to use for resuspending the powder. The bottle states that it is soluble in water.

Has anyone used cellulase for a similar application? What is the optimal concentration, pH, temperature, and buffer for cellulase enzymatic activity? Any recommendations would be appreciated!
