OPA degree of hydrolysis?

Hi there, I am having trouble calculating the degree of hydrolysis for my protein samples. I have obtained the absorbance, blank and serine standard readings, but the problem I am having is where the alpha and beta values fit into the calculation? Are these just taking simply as the calculated serine value minus beta, all divided by alpha, or is there more to it than that? thanks

That seems to be right according to what you are stating. I do something very similar in my calculations, the original Nielsen article describes it well. But it is not an overly difficult calculation, no. Not sure that you have it but the original paper is linked here.
