Now i am going to do antibacterial activity using crude plant extract so for that how do i make different concentration of 10,20,30ug/ml from stock 1mg/mL. but to my knowledge i am using following formula is, C1v1=c2v2, my initial concentration is 1mg/mL from these i have prepared above mentioned concentrations that 10ug*100ul/1mg so i got 1ul plus 99 ul dmso= 10ug/ml so this is crt or not, please clear my doubt...
The c1v1=c2v2 theory is correct.
1mg/mL = 1ug/uL by definition. So, a 1mL solution of a 10ug/mL stock would be made of 10uL of 1mg/mL stock and 990uL of DMSO. In the case that you wish to end up with less, you simply convert this. The same can be applied to the other concentrations.
Getting accurate measurements from very small volumes like this are a quantitative nightmare. Most of the time, a serial dilution is the recommended step, so that minor variations in your pipetting don't destroy your concentrations. Especially when working with DMSO, the viscosity can be an issue, so ensure that you "prime" your pipette correctly, hold it directly vertically, and pipette with care. Also, be sure to mix each step of the serial dilution before pipetting to the next gradient vial (usually cycle the contents 3x).

First of all,obtain the MIC of this crude by serially dilution of your stock( 1mg/mL ) or 1000microlitre.Prepare 10 sterile eppendorfs in each one 50 microlitre sterile distilled water.Add 50microlitre to the first one(0.5mg/ml) and from this one after mixing transfere 50to the second(0.25 mg/mL) and so on and make the assay.
I guess the answers so far are quite illustrative. However, it might be less confusing to do your conversions before dilution so you could work at the same unit (e.g dilutions of mg/mL to mg/mL). Also, since you are working with crude extract, you should try and avoid having solid plant materials in your stock-solution (before dilution) through proper filtration; else, the dilution concentration would not reflect the corresponding calculated values for your benchwork deductions.
