1. To create micronutrient deficiency: Don't give the plant any micronutrients. As you imply, sand is a good way to start as there isn't much additional nutrition in the form of organic materials or clay minerals. Wash the sand with deionised water several times, maybe even with 1% nitric acid to leach out any free elements (vs bound elements in the soil particles).
2. EDTA is probably not going to bind up the Zn and Fe sufficiently before it is broken down by the soil microbiota. You might be able to limit the free Zn or Fe for a little while, but plants have various root exudates that will compete with the EDTA and the plant will probably outlast the chemical.
3. How much concentration of what and for what purpose? If you are trying to induce a deficiency, don't add anything at all. If you want to guarantee sufficiency, check what other researchers have used on your particular crop. You didn't tell us what species or soil or any other conditions, so we can't really advise on that.
4. You might get an antagonistic reaction with P, but you might equally have a fertilizer with a sufficient amount of trace element that it doesn't react. Plus you no longer have a "simple" deficiency, but an "induced deficiency" caused by luxury amounts of a second element. Is that what your study is investigating?
Arid soils are already readymade soils extensively deficient in Fe and Zn ..??
