This was isolated from a 1000 year old Quercus robur oak tissue culture. The colonies on LB low salt agar plates look glossy, white-ish, smooth edges. They grow very slowly. In a month at room temperature they grew what E.coli can grow in less than overnight. The black and white photo is 10 000x magnification, the other one is Gram staining at 1000x magnification (The Gram stain might be done poorly). Can anyone help me with identifying this? I'm hoping it's an endophyte, potentially yeast, but it might be a pathogen of some sort.
The best way to fully identify is 16S rRNA
U can send the culture to specific lab in DNA isolation then PCR product and finally sequencing
From the sequencing go to NCBI database to do the phylogentic tree

Measure the size of organisms using micrometry and proceed with 16S rRNA sequencing.
The best way is to do DNA sequencing.
There are many studies that identified the presence of Saccharomyces n Candida species on old oak trees. Grow them in Yeast specific growth media like, YPD or Sabbaroud Dextrose agar to check growth rates and cell morphology. Chromagar or BIGGY'S agar and sugar fermentation tests for yeasts may allow presumptive identification.
Yet DNA sequencing ( especially ITS region)is the best for confirmation.
oval shape and budding in large cells (nuclear division) make me thought it species from yeast.
Based on the microscopic morphology, it gives resemblance to yeast. You should try to culture your isolate on mycological media, such as Sabouraud dextrose agar, Malt extract, Pal sunflower seed medium etc., and study detailed microscopic morphology in Narayan stain or PHOL stain or Lactophenol cotton blue stain. If possible, you can go for molecular technique for the identification.
if the magnification is correct regarding the image you give, the b & w picture gives about 1 µm for the cell dimension. This is too small for a yeast cell!
