I'm making nano/microbubbles with 2 different compositions.

1. Combination of phopholipids, cholesterol, and DSPE-PEG2K

2. PLGA+cholesterol

Both of them I do with double emulsion method followed by sonication (ice bath sonication). The first emulsion, i mix 5:1 solvent (chloroform: water) then used PVA 4% to form the second emulsion.

I separate the results from each composition to get the particles through freeze-drying and centrifuge.

1. The freeze-dry result i dilute with water

2. The pellet from centrifuge i also dilute with water

My problem is all of them (particles) can't dissolve in water.

Are there any mistakes in the way I work? or something missing?

It is also my first time to synthesize nanoparticles, i'm afraid i did wrong all the whole time
