Hey all!
I am attempting to create PLGA microspheres using ammonium bicarbonate in the dissolved PLGA + dichloromethane solution, then added to oil with a homogenizer. However, every time I add the PLGA+ dichloro to the oil, it precipitates around the homogenizer and doesn't ever make the small spheres I'm looking for, even without the ammonium bicarbonate added.
Any solutions?

For accurate answer, we need to know:
1. what oil you are trying to disperse microspheres in and in what concentration?
2. is it only oil or o /w mixture?
It is just vegetable oil and at a 70:30 ratio oil to PLGA
It is oil then added to it is the PLGA+Dichloro mixture
It happens no matter the amount of oil but the methods I was using called for that ratio.
Try a milder technique instead of homogenization.
What about an over-head stirrer set at e.g. 500 rpm (or perhaps 1000 rpm, if that doesn't work)?
