You might want to give Accutase a try it's gentler than trypsin and often better for loosening monocytes without harming them.

Alternatively, an enzyme-free dissociation buffer could work well since it's designed to gently detach cells without affecting surface markers.

Lastly, you could also try incubating with EDTA in PBS for 10 to 15 minutes at 37°C this can help break some of the adhesion.
