Hi,

If you want to silence a single miRNA, I recommend you to use the technology based on antagomirs, that has great results. However, if you are looking for the complete depletion of miRNA the best shot would be to silence the expression of either Dicer or Ago1.

Hi, I think you can transfect a miRNA-sponge with selection marker.

Guangyun Tan, great idea of the sponge! Much more suitable for my purposes.

In a meantime I have found another solution: miRCURY LNA by Exiqon.

The point is if you are looking forward to downregulate the miRNA. Its always suggested to go for Antigomirs with 2'O me modifications which shows best inhibitor response to endogeous miRNA. The idea of LNA inhibitor is also good. The another alternative is Dharmacon miRNA hairpin inhibitors.

It works well and is cost effective.
