Anyone experience MHCI tetramers binding to CD4+ Tcells? In some samples I have more tetramer positive CD4 T cells than CD8 T cells. I'm worried that if I titrate the tetramer enough to get rid of CD4 background then I may lose my CD8 signal as well. Is there an obvious answer I'm missing? Maybe my CD4 antibody itself is binding the tetramer?
Dear Nancy, in mice there is significant percent of MHC class II reacting TCRs in CD8 population, but not vice versa. So, I would consider your effect as unusual. It may be, your observation is related to impairment of intrathymic selection and autoimmune disorders in patients. Also, you need to rule out possible artifacts caused by the aggregation of cells.
