Please share your ideas on possible methods of free amino acid extraction. Normally we use dil HCl for extraction. I wish to extract from other medium. Can anyone suggest what are the other methods can be employed to get a better yield? Such as using enzyme, supercritical, microwave assisted extraction etc. Do anyone has any experience in this?
I would like to ask you what is the goal of your extraction. Is it to recover the total proteins content, to extract just free amino acids or some specific protein? Depending on that the technique used could be diferent.

As you said previously the sue of diluted HCl has been proved to be a good method for amino acid extraction. After that precipitation of protein by means of trichloroacetic acid or ethanol can be used. We used this method to extract amino acids and short peptides from meat and cured ham.

Actually, HCl extracts amino acid, peptides and a lot of proteins from the matrix, so a further step of de-proteinization has to be done using ethanol or trichloroacetic acid. I am afraid that that one are the only method I have been used.
I guess if you can use ethanol or trichloroacetic directly in the raw material for this extraction. I imagine that any other acid as formic, acetic, sulphuric or nitric could give same results. But I never tried before and I could be wrong.

what is the concentration of HCl, temperature and time you have used for the extraction?
try to do acid hydrolysis with 6 M HCl at 110°C for 23 hours. Amino acids containing sulphur is hydrolysed separately with 6 M HCl after oxidizing (formic acid + hydrogen peroxide, 9:1 v/v, 20 h at 4±1°C).
many methods are used in extraction of amino acids, but must define your goal from extraction. you can use HCl hydrolysis, ethanol, chlorophorm etc. Firstly define your goal and in what this extract will be used.
