I am starting a membrane vesicle assay shortly. As I am doing it for the first time I don't know which filter to use for rapid filtration. The source of vesicles is HEK293 & CHO Cells. Literature mentions 0.45um pore size. But I am confused with the material used for filtration. For insect cells derived vesicles, people use Glass Fibre filters of 1um pore size but glass fibre filters are not available for 0.45um pore size specification, instead made of PVDF (Hydrophilic). Should I go with PVDF?

You not only need small pores but also need as thin as the possible membrane for you vesicle filtering.
I used the nanosieve membranes from Aquamarijn, and it worked quite Ok. It is LPCVD SiN filter, with 450 nm pores, the thickness of ca. 500 nm. You can do surface functionalization before filtering as well.
