i did mechanical compression on cancer cells and then looked at a-tubulin staining pattern. I lack experience and did not see any changes in a-tubulin pattern.
To my opinion, the upper cells seem more epithelial-like, the other cells (uncompressed) are more mesenchymal. alpha tubulin pattern looks changed. You most probably damaged the integrity of cell skeleton structure. Because the most of the cells lost their membrane and look unhealthy in the compressed cell image.
I think, In the compressed cells, tubulin proteins were smashed, if there was an intact membrane, they should have stayed organized like uncompressed cells. you can test that just adding crystal violet on the cells without fixation. If the membrane is intact, crystal violet can not enter the healthy cells.
