In our experimentation, end point is quantification of NK cells after treatment period. Different markers like CD25, CD69, CD16 etc., were given In literature (there is no uniqueness in marker selection for quantification of NK cells). So, here my basic doubt is, what is basis / rationale to select the marker for the quantification of NK cells and what is ideal marker for the same?
So... I went with the easy marker, CD161 (the human equivalent of NK1.1). I gate on CD3- cells C161+ in the lymphocyte gate.
Clinically, the number of NK cells in blood are usually determined using a combination of anti-CD16 and -CD56 in the same channel. This will pick up both CD16+/CD56low and CD16-CD56high cells, and usually all cells that lack expression of CD3 in the lymphocyte gate but express any of these markers are considered to be NK cells. If you have the ability for multicolour analysis you can of course use them in different channels to divide them into the subpopulations instead.
