Contrasting parents for sucking pest were crossed. There were 4 crosses. Now, there are a very few advanced generation (F8) progenies in each of the 4 crosses, about 8-10 in each. Within the progenies, there are distinct differences for the pest reaction. Can we use them for mapping? How exactly?
Hi sir, You can screen the advanced generation progenies tolerant to different pests using markers and identify the desirable one. But these advanced generations (F 8) may not be suitable for mapping population.
Dear Sir, If the aim of gene mapping is to identify markers linked to the QTL that is associated with sucking pest tolerance, for their utilization in MAS. Phenotying and genotyping F8 progenies from all the crosses together with suitable molecular markers and subjecting the data for one way ANOVA for marker trait association analysis (Single Marker Analysis) may help you to find putative markers linked to sucking pest tolerance.
This is my opinion and may the experts can give better suggestions and correct me if i am vague.
Thanks A.B.M Sirisha and Venkatesh Raichur for your suggestions. I also had this in mind. Why not do BSA with these F8 progenies in each of the 4 crosses? Now that the progenies have attained a fair bit of homozygosity. We can always validate it in F8-9 progenies later.
Another thing which is less addressed is the involvement of the Biochemical basis of tolerance/resistance. If you can get and share more resources on this aspect, please share.
