For Malaria Rapid test, we use whole blood as sample for diagnosis of Malaria. It required almost 20-25 minute to clear reddish back ground from nitrocellulose membrane. What should we do to clear background within 10 minute, without reducing sensitivity and specificity.

,We have project to develop test, we are not looking for finish test.
Try membrane with different flow rate,check the sample or wash buffer volume or try other detergent, different detergent concentration.
I agree with Oded . I think you have to modify the variables which affect the rate of migration .good luck

I had try with wicking rate membrane, but problem not resolve.
For wash buffer we had try Saponon, Brij, Tween, Triton, Tergitol.
Did you add blocker to the membrane?
You can replace the sample application pad to one with better separation for the red blood cells.

For malaria we have to lyse RBC, i think hemoglobin is not releasing from NCM.

The detergent you are using are ok. Maybee higher precentage. I still suspect the membrane . Try membrane like HF80, 120,140 check the rate and flow. Too quick add PEG

Test has limitation for buffer volume,we can add more then 0.100 ml to 0.120 ml
The slow capillary rate in your LFIA is due to blocking of membrane by BSA or Casein. Donot block the membrane. First treat your sample pad with the detergent before assembling the all component of the LFIA. Now on treated sample pad add the sample (blood) it will lyse the RBC.

Use membrane of pore size higher than 10 micron. Thats it. And also
You can try wash buffer, gold conjugate or blocking buffer from IRIS NANOTECH. May be it can solve the problem.
