I am currently investigating TAA-based immunotherapy using the MC-38 colon carcinoma mouse model and have encountered an unexpected observation during my experiments. Specifically, when culturing splenocytes isolated from tumor-bearing mice overnight with tumor-associated antigen to evaluate TH1-specific responses (particularly IFNγ and TNFα production), I consistently observe a notable reduction in CD8+ T cell numbers.
I have encountered this phenomenon on two separate occasions. Could anyone with experience in tumor immunotherapy studies clarify whether this loss of CD8+ cells is a commonly observed phenomenon, or if it might indicate a potential issue with my experimental protocol?
