I've already been using a combination of poly-d-lysine, poly-ornithine, laminin, and fibronectin but am still having issues with adherence of a tricky disease line. Any thoughts?
Have you tried coated plastic coverslips? iPS cells usually adhere easily on plastic cover slips coated with either laminin or poly-l-lysine. Make sure that you give them several days to settle down.
I used poly-L-ornithine 2h in the incubator, then you remove it and after that, you coat with matrigel for 1 hour (or more) at room temperature, that you will remove right before plating motoneurons, it works well.
