I have a question, does anybody know how to stain living cells for several days? Most of stains are toxic for cells or require fixations. We need the stained cells alaive for at least 3 days.

Is this what purpose and which cells are you using?
Generally lymphocytes can be stained with cell proliferation dyes (Eg. CFSE) to monitor their proliferation for up to 7 days.

@ Amarendra: It is of general interest. One project is cell migration the other just to identify cells in a matrix. I´ll take every protocol which might help :o)

In my experience, I could observe living cells with Calcein-AM in DMEM (without phenol red) for around 100 hours.
