I had performed ligation reaction at 4 °C (overnight) and 23 °C (4 hours) and when i transformed my ligation mixture using DH5-alpha competent cells, colonies appeared only with 4°C ligation mixture but not one with 23°C. But the 4 °C mixture gave false positive colonies (self ligation of vector) when screened. How to tackle this issue? Any valuable suggestion will be highly appreciated.
