How do I calculate the enzyme activity of laccase if I have got an OD of 0.069 when I put the laccase assay using ABTS as a substrate?

You have to measure the change in absorbance over time, i.e. you have to measure at different time points and then calculate the ΔA/min by linear regression. Please refer to the following discussion on how to calculate the activity once you have determined ΔA/min.
