I am currently screening heat stress tolerant Lablab purpureus. One of the method is to quantitatively determine superoxide anion by using NBT staining method. I immersed the leaves in NBT dye for 8 hours, and was trying to remove chlorophyll by boiling the leaves in 80 celcius alcohol. It took me about half an hour to only remove the chlorophyll partially. Any suggestions how to remove it rapidly? Thank you.
