Dear all,
I want to perform a KI67 staining on 3D tissue-engineered skeletal muscle of approximately 2cm long and 3mm thick (cilindrical shape). Does anyone already performed a 3D staining with KI67 on tissue with comparable dimensions and if so can I have some tips or working protocol?
I would combine the KI67 staining with a clearing as well (this clearing has already been established in the lab).

An antibody will not diffuse through a tissue block of those dimensions. An antibody-based staining protocol only works well on thin (i.e., 5-10 µm) sections.
Gordon L Warren First of all thank you for your reply. I found this protocol on jove.com (performed on whole mount skin preparations) that's why I wanted to give it a try. However, I agree with your answer and wanted to check whether others got some experience with it or could provide me with some tips.
Do you think this protocol is worth the try or what is your experience with 3D whole mount staning?

Mouse skin is considerably thinner than a 3-mm diameter cylinder of muscle. Mouse skin thickness is in the 0.5-0.8 mm range.
