I want to distinguish kupffer cells (KC) from human primary hepatocytes (PHH) . is there any specific marker expressed by activated/inactivated KC but not by PHH?
I would try dual staining with CD45 and CD163 or CD206. CD45 is expressed by KC and not hepatocytes. Soluble versions of CD163 and CD206 are markers of KC activation and these markers are also expressed/upregulated on KC.
I've detected some FcgRs mRNA in the KC-hepatocyte co-culture system. According to the literature, FcgRs express on phagocytes involving DCs, macrophages, monocytes and so on. Can I use FcgRs as KC-specific marker? Does hepatocyte express FcgR genes?
I'm not aware of any FcGR expression by hepatocytes. However, your hepatocyte/KC preparation may contain some liver sinusoidal endothelial cells. These LSECs have very similar antigenic profiles as KC, including expression of CD16 and CD32 (no CD64). CD45 and HLA-DR are highly expressed by KC and not LSECs or hepatocytes (see linked paper). I use CD14 and CD45 for KC, but CD14 is also expressed on hepatocytes, so you need the combination. I have not seen high-level CD64 on KC, so I think CD14 is a brighter, more distinctive marker.
