Non-adherent M-CSF differentiated bone marrow-derived cells were harvested on day five of culture by gentle pipetting and were subsequently subjected to MACS-mediated depletion of CD117 + stem- and progenitor cells. Cell suspensions harvested under these conditions were verified to consist of 86±6% mature monocytes (NOT MACROPHAGES) by means of FACS analysis.

I used to isolate mouse and human neutrophils in the lab. The process should be the same- spin it down and collect the cells from the proper band. If I recall correctly, monocytes and lymphocytes are in band 3.
