I have prepared slides for histological studies. I want to do SEM for that specimens. Can same slides would be used after some treatments???? If yes what kind of treatment should I use for this. Secondly I had preserved tissues with me embedding on wax can that can be cut with ultramicrotome and used as it for SEM????
Short answer - no.
With SEM you see surface of a sample, in your case it will be the surface of coverslip, not your section.
There are some methods for salvaging such sections, but they usually give not very good results. It is much better to prepare separate specimens for electron microscopy.
By the way, SEM is not really good for histology, you may want to use TEM instead.
No you can not use the same slides. You need to prepare SEM specimen on specific stubs. Moreover SEM is just for surface analysis. You should use TEM for internal cell analysis. Prepare your samples as per requirements of TEM.
