(Felczykowska et al., 2014): Overnight cultures from E. coli were centrifuged at 3,500 rpm for 10 min. Afterwards, the cell pellets were re-suspended in 0.01 M Tris-HCl pH 7. Finally the extracts were sonicated at 20% maximal amplitude for 370 sec (10 sec with 10 sec intervals without sonication), on ice and the extracts were sterile-filtered with 0.2 µm membrane filters (Corning).
So my question is, will the extract be a protein extract? (since we take the cell pellet only), or a total extract? (including proteins and secondary metabolites), also, will it exclude those secreted outside of the cell?
