We are trying to isolate total RNA from pomegranate leaves, we tried using several kits but purity was the problem and it requires an additional manual purification for better purity. However there is loss in the quantity during purification process, are there any kits/manual methods to isolate total RNA from pomegranate leaves?
Please assist me in this regard

i think, you are taking huge amount of sample or crushing is not proper, you can use any method or kit but mind these two crucial things.

it is recommended using TRIzol reagent. it is the best kit to isolate rna.

Thank you for reply i tried using 100mg of sample i didn't get anyat first , i tried M&N, Sigma kits, Trizol but i didnt get in any. However we did little Modification with Buffers and Binding solution now we are getting good QUALITY but not good Quantity.
