Callus from anther is achieved on MS+1-2mg/L 2,4-D

Regeneration medium (MS) should contain BA (1-2mg/L)+GA3 (0.5-1mg/l)

We got callus from anther on MS medium supplemented with 2,4-D (8micromolar). Other options are Nitsch and Nitsch medium and Whites medium

I tried MS media with NAA and BAP at higher concentrations i.e 5ml/L each, got callus, but they showed very poor response on sub culturing, wat do you suggest? wat combinations should i use for regeneration? Nitch and Nitch media and N6 media are same or not? ready made whites media is available or not?

nitch & nitch meium and N6 medium are not the same. In barley AC we omit NAA & BAP in the regeneration media, perhaps you should omit those in your regeneration media and see if the regeneration frequency increases.
