If agar well method is not working then, you may try Kirby-Bauer disc diffusion method.

I agree with Tambat, I have use disc diffusion with antifungal activity

what are you working on? Mycelial fungi, Candida ...?

Thank you all. @Salam and @Rushikesh please let me know did you people applied your concentrations in the wells or on discs first and then kept the fungus for growth or firstly spread the fungal spores as like bacteria and then made the wells?

Instead of spreading your fungal culture on plate, you should make a seed plate agar having desired CFU of your culture and then pour it in Petri dish. Allow the plate to solidify and then make a well or disc whatever it is. Then keep the plates in refrigeration for 1-2 hours to allow the content in the wells diffuse freely through the agar. And then incubate it.

I think you should follow Tambats suggestion -

Liquid culture method may also be better for antifungal activities than the agar process- I find tit more convinient.

We usually use broth dilution method for antifungal susceptibility testing according to CLSI documents.

You can use CLSI M38-A2 protocol witch is standardized for filamentous fungi.
