I am working on an experiment using Ulva lactuca as a substrate. At first I place 5 grams in 100 ml 0.4 N HCl .After this I added cellulase the reducing sugars rose to about 34 mg/ml. After getting this result I increased to volume to 1 L but the same concentration as before 5% (50 g in 1000 L) the concentration of reducing sugars changed to 136 mg/ml. I am not sure if this is accurate as I believe the concentration should be same as before around 34 mg/ml. Really looking forward to your response.
Did you add the same enzyme concentration?, check the enzyme concentration. Secondly did you check pH and temperature?. How long did you incubate in both experiments?. It's very important to mantain these conditions exactly the same, otherwise the results would change dramatically. Lastly did you consider the total volumen in the calculation sheet?. If you still have problems after this, then check the substrate because you are working with a non-purified substrate, thus homogenity is very important. Good luck
Hello, have you kept the relationship between the concentration of substrate and the enzyme constant? Are the hydrolysis conditions exactly the same? pH, agitation, ... etc?
