Hi . Is there any way to use EDC/NHS coupling system for histidine without oligomerization of itself. Actually I want it to react with amine groups of another substance . what is the appropriate medium for this reaction ? Is it possible to use two medium, one for EDC/NHS and histidine with lower pH and another medium that has higher pH for another substance?
I'm unfamiliar with biphasic reaction media for amide couplings using EDC/NHS, (the Schotten-Baumann does exist however https://en.wikipedia.org/wiki/Schotten%E2%80%93Baumann_reaction) but you could do a couple of things:
First protect the nitrogens of your histidine using trityl for the imidazole moiety and Fmoc/Cbz/Boc for the amine, then proceed with your coupling.
I did coupling in water for proteins to nanoparticles, and it worked. I think a neutral buffer should be fine too. It is not the optimal condition for coupling, but it is not that far off.
