Salinity influence the microorganism (seed) activity in the saline water during incubation, if microbial population decreased; will it affect the results?
For a correct BOD analysis you should use a "seed" which is adapted to the actual salinity. The principle is to measure the biodegradability of the sample - not the toxicity.
I agree with Henrik and would add that a good place to obtain such adapted seed material is water from the receiving body downstream of the point of discharge. If that is unavailable, an acclimated seed can be created in the lab. Check Standard Methods for the Examination of Water and Wastewater 22nd edition, section 5-6.
I agree all of you and I recommend that seeding matter is the key factor for BOD determination. you can also make a calibration curve for BOD and Salinity, it can help you for relationship between salinity and detection limit of BOD in seawater.
