I guess the first problem is quantification of muscle myostatin levels. Western blot's will give you change in expression, but not inter-individual comparisons between gels, meaning you could only examine a small cohort.
If someone has reported a standardized protocol for extraction of total protein from individuals muscle biopsies, AND then true quantification by ELISA, and a comparison of this result by to serum myostatin by ELISA, that would give the answer you're looking for. I'm not aware of this in the literature however.
