The cryostats in our lab is not delivering sections as expected. it seems like whatever section thickness we choose (e.g. 20, 30, 40, 60 microns), the real thickness we get are around 20 microns always. Did anybody face this problem before? I am stuck here with all my research works due to this one problem. I am in need of help. Thanks

Have you tried to reset everything? Potentially defrosting the cryostat, cleaning everything and restarting it? It can help if it's not sectioning as it should. Otherwise, if you want to try another equipment, you could try to section the brain with a microtome. But you will get floating slices in this case.
